GHRP-1 vs GHRP-6
The prototype versus the one that actually got studied. Both are early ghrelin-receptor peptides; GHRP-6 has far more human pharmacology and a defining appetite effect.
Short answer
GHRP-1 and GHRP-6 both have human clinical data, so the deciding factor is what you want them for. GHRP-1 suits someone focused on GH boost, while GHRP-6 is aimed at muscle and recovery. For GHRP-1, a single IV dose raised GH, peaking at 15 to 30 minutes, in 15 short children (1993); no outcome trials.
GHRP-1
The original GH-releasing peptide: a dose genuinely triggers a GH pulse, but that pulse is a surrogate marker — not a proven body-composition or longevity outcome — and it is unapproved.

Pulses GH short-term, but its most reliable real-world effect is a surge in appetite; no long-term human outcomes and not approved.

marks a row where the two differ. “Not established” marks a cell the available evidence does not answer. Evidence grades come from our evidence matrix, which grades each molecule on the human data for the use it is marketed for.

Our verdict
GHRP-1 does what its class does: an intravenous bolus produced a progressive rise in plasma GH peaking within about 15 to 30 minutes in healthy children and adolescents, with a larger response in pubertal than prepubertal subjects. GHRP-6 has broader human characterization, with GH release documented across intravenous, subcutaneous, intranasal and oral routes, plus a strong dose-dependent appetite effect and a measurable rise in ACTH and cortisol. Both stop at the surrogate marker; there are no long-term human outcome trials behind the fat-loss, muscle or anti-aging claims for either. Neither is approved, both are research-only, and both are prohibited in sport.
GHRP-1 fits if
GHRP-1 is mostly of historical interest, relevant to a reader tracing where this peptide family started; almost everything specific about GHRP dosing and side effects comes from its descendants.
GHRP-6 fits if
GHRP-6 is the more informative read, since its human pharmacology is broader and its appetite effect is the clearest signal it reliably produces.
GHRP-1 vs GHRP-6: common questions
- Is either GHRP-1 or GHRP-6 FDA-approved?
- No. GHRP-1 is not FDA-approved; sold as a gray-market research peptide; prohibited in sport. GHRP-6 is not FDA-approved; sold as a gray-market research peptide.
- Which has stronger human evidence, GHRP-1 or GHRP-6?
- Neither clearly. Both have human clinical data. The key source for GHRP-1 is Laron, Acta Endocrinol 1993; for GHRP-6, it is Jaffe, JCEM 1993.
- What are the main safety concerns with GHRP-1 and GHRP-6?
- GHRP-1 has no long-term human safety data; expected appetite, cortisol and prolactin effects. For GHRP-6, watch for strong appetite increase; raises ACTH and cortisol; GH response blunts with repeated dosing.

The full evidence reviews
- GHRP-1: the original GH secretagogue, and why a real GH pulse still isn't proof
GHRP-1 is one of the earliest synthetic growth-hormone-releasing peptides and the forerunner of GHRP-2 and GHRP-6. A single dose genuinely releases growth hormone — but that pulse is a short-term surrogate marker, not an outcome, and there are no long-term human trials behind its anti-aging marketing.

- GHRP-6: the 'hunger' growth-hormone secretagogue, and what the evidence shows
GHRP-6 is a first-generation ghrelin-receptor agonist that pulses growth hormone — but its most reliable real-world effect is a strong, dose-dependent surge in appetite. Real short-term GH pharmacology, preclinical cytoprotection, no long-term human outcomes, and not FDA-approved.
